The publication of Can-Fite BioPharma's Phase III clinical trial results in a high-impact journal signifies a pivotal moment for the company's drug candidate, Piclidenoson. The drug's potential to treat moderate to severe plaque psoriasis with a favorable safety and efficacy profile could disrupt the current market, which is dominated by biologics and topical treatments. The reported PASI-75 rate, a standard benchmark for psoriasis treatments, indicates a statistically significant improvement over placebo. However, the rate itself may be compared to existing therapies to assess competitiveness.

Investors should monitor the progress of the ongoing COMFORT-2 trial, as its outcomes will be critical for FDA and EMA approval decisions. Success in this trial could lead to market entry and potential partnerships or acquisition interest. Conversely, any setbacks could significantly impact Can-Fite's valuation and future prospects.

From a financial perspective, the development stage of Piclidenoson is a capital-intensive phase. The company's ability to manage costs and secure additional funding, if necessary, will be crucial for sustaining operations and reaching commercialization.

The endorsement of Piclidenoson by a key opinion leader such as Dr. K.A Papp adds credibility to the drug's profile. In dermatology, the therapeutic landscape for psoriasis is competitive, with patients and providers seeking treatments that balance efficacy with minimal side effects. Oral administration of Piclidenoson is a notable advantage, potentially improving patient compliance compared to injectables.

Long-term safety is paramount in chronic conditions like psoriasis; thus, Piclidenoson's tolerability is a significant factor. The drug's linear increase in efficacy over time suggests a sustained therapeutic effect, which is critical for chronic management. However, it is essential to compare these results with long-term data from competitors to fully understand Piclidenoson's market position.

The progression of Piclidenoson to a pivotal Phase III study is a noteworthy milestone in Can-Fite's clinical development pipeline. The drug's mechanism of action as a small molecule could offer a different approach to psoriasis treatment compared to existing therapies. The significance of the trial's approval by both the FDA and EMA cannot be overstated, as it implies a recognition of the drug's potential and a rigorous review process.

Investors should consider the broader implications of Piclidenoson's success on Can-Fite's pipeline. A positive outcome could validate the company's platform technology and bolster the development of its other drug candidates. Conversely, any adverse findings could have a ripple effect across the company's portfolio.

Piclidenoson is a small molecule safe oral drug suitable for the chronic treatment of patients who suffer from moderate to severe plaque psoriasis.

PETACH TIKVA, Israel--(BUSINESS WIRE)-- Can-Fite BioPharma Ltd. (NYSE American: CANF) (TASE: CANF), a biotechnology company advancing a pipeline of proprietary small molecule drugs that address oncological and inflammatory diseases, today announced that the Journal of the European Academy of Dermatology and Venereology (EADV) published an article titled “Efficacy and safety of piclidenoson in plaque psoriasis: Results from a randomized phase 3 clinical trial (COMFORT-1)”. EADV is a top ranked peer reviewed journal (impact factor 9.2) that publishes articles on clinical and basic science topics in dermatology. Article Link.

The article’s first author, Dr. K.A Papp, is an internationally renowned key opinion leader in the psoriasis field and was the engine for some registered drugs on the market for this devastating skin disease.

The EADV article presents the safety and efficacy of Piclidenoson in the randomized, placebo- and active-controlled, double-blind Phase III COMFORT-1 trial. As previously reported, the study met its primary endpoint which was the proportion of patients achieving ≥75% improvement in Psoriasis Area and Severity Index (PASI) from baseline (PASI-75) at Week 16 (3 mg BID dose: PASI 75 rate of 9.7% vs. 2.6% for Piclidenoson vs. placebo, p=0.037). Piclidenoson’s efficacy continued to increase throughout the study period in a linear manner with an excellent safety and tolerability profile.

Currently, Piclidenoson is being evaluated in COMFORT-2 a pivotal Phase III study that has been approved by both the U.S. Food and Drug Administration (FDA) and the European Medicines Agency (EMA).

“We are very much encouraged by the excellent data in the first Phase III study demonstrating the significant effect of Piclidenoson in psoriasis,” stated Dr. Fishman, Can-Fite’s Chief Scientific Officer and Executive Chairman. “We plan to start treating patients in our pivotal trial very shortly and hope that Piclidenoson, with its safety and efficacy profile, given orally to psoriasis patients will become a drug of choice.”
