Rhea-AI Filing Summary

iBio, Inc. reported preclinical results from an eight-week obese non-human primate study of IBIO-610, an Activin E antibody, alone and with semaglutide. Lean mass accounted for 6% of weight lost with the combination, compared with 22% with semaglutide alone. At week 8, fat accounted for 93% and 77% of weight lost, respectively.

After a single intravenous dose, IBIO-610 showed a 97% reduction in serum levels of active Activin E at week 8 and had a 33.2-day half-life in NHPs. iBio said a projected human half-life could potentially support twice-yearly dosing and plans to begin first-in-human studies in the first half of 2027. The company noted that NHP findings may not be replicated in humans.

Reduction in serum levels of active Activin E 97% At week 8 following a single intravenous dose of IBIO-610 in NHPs
Half-life 33.2 days IBIO-610 in NHPs
Study group size 6 NHPs per group Eight-week obese NHP study
Planned first-in-human studies First half of 2027 iBio's stated plan
Activin E antibody medical
"IBIO-610, an Activin E antibody"
GLP-1 receptor agonist medical
"semaglutide, a GLP-1 receptor agonist used to treat obesity"
A GLP-1 receptor agonist is a medicine that mimics a natural gut hormone to trigger insulin release, slow stomach emptying, and curb appetite — like using a key to turn on a lock that controls blood sugar and hunger signals. For investors, these drugs matter because they treat common conditions such as diabetes and obesity, can drive large prescription and sales growth, reshape healthcare costs, and heavily affect drug pipelines, competition and company valuations.
non-human primate (NHP) medical
"obese non-human primate (NHP) study"
Non-human primate (NHP) refers to monkeys, apes or other primate species used in medical research to test safety and effectiveness before treatments are given to people. Because their biology is closer to humans than other lab animals, results from NHP studies can strongly influence whether regulators allow human trials, how quickly a drug advances, and the cost, timing and risk profile investors use to value biotech projects — like a dress rehearsal that helps predict real-world performance.
serum levels of active Activin E technical
"a 97% reduction in serum levels of active Activin E"
first-in-human studies medical
"plans to begin first-in-human studies"
A first-in-human study is the initial clinical trial where a new drug, biologic, or medical device is tested in people for the first time, after laboratory and animal testing. It focuses on safety, tolerability and basic dosing rather than proving effectiveness; think of it as the first road test for a prototype. Results matter to investors because they provide early human data that changes the perceived scientific risk, timeline and potential value of a development program.

Item 7.01. Regulation FD Disclosure.

On October 13, 2026, iBio, Inc. (the “Company”) issued a press release highlighting preclinical body-composition data from its eight week obese non-human primate (“NHP”) study evaluating IBIO-610, potentially a first-in-class Activin E antibody candidate. The data were discussed in an oral presentation at the European Association for the Study of Diabetes Annual Meeting on October 1, 2026.

In this NHP study, lean mass made up 6% of weight loss with IBIO-610 plus semaglutide, verses 22% with semaglutide alone and IBIO-610 delivered durable, near-complete inhibition of active Activin E, and when combined with semaglutide, nearly all observed weight loss came from fat. Additional analysis of the eight-week obese NHP study (n=6 per group) further characterized the potential for IBIO-610 to support muscle preservation when combined with semaglutide. At week 8, fat accounted for 93% of the weight lost by NHPs given a single dose of IBIO-610 alongside twice-weekly semaglutide, compared with 77% for NHPs given semaglutide alone.

These results build on previously reported data from this NHP study, in which IBIO-610 demonstrated a 97% reduction in serum levels of active Activin E at week 8 following a single intravenous dose. The study also found that IBIO-610 had a 33.2-day half-life in NHPs, with a projected human half-life potentially supporting twice-yearly dosing. The Company plans to begin first-in-human studies in the first half of 2027.

iBio Highlights Data from a Non-human Primate Study Supporting IBIO-610’s Potential to Preserve Muscle in Combination with GLP-1 Therapy

In an eight-week obese NHP study, lean mass made up 6% of weight lost with IBIO-610, an Activin E antibody, plus semaglutide, versus 22% with semaglutide alone

iBio expects to start first-in-human trials of IBIO-610 in first half of 2027

SAN DIEGO, Oct. 13, 2026 -- iBio, Inc. (NASDAQ: IBIO), a clinical-stage biotechnology company developing long-acting antibody therapeutics for obesity, cardiometabolic and cardiopulmonary diseases, highlighted preclinical body-composition data from its obese non-human primate (NHP) study evaluating IBIO-610, potentially a first-in-class Activin E antibody candidate, alone and in combination with semaglutide, a GLP-1 receptor agonist used to treat obesity. The data were discussed in an oral presentation at the European Association for the Study of Diabetes (EASD) Annual Meeting on October 1, 2026.

“While GLP-1 therapies have transformed obesity care, patients still need additional options to improve the quality of weight loss,” said Martin Brenner, DVM, Ph.D., Chief Executive Officer and Chief Scientific Officer of iBio. “In this NHP study, IBIO-610 delivered durable, near-complete inhibition of active Activin E, and when combined with semaglutide, nearly all observed weight loss came from fat. We plan to begin first-in-human studies in the first half of 2027. Our focus is on translating this biology into a potential new therapeutic option for patients living with obesity and cardiometabolic disease.”
