Rhea-AI Filing Summary

First Tracks Biotherapeutics, Inc. (TRAX) reported positive top-line results from Cohort 1 of its Phase 1b ANB033 celiac disease trial. The 32 patients who completed dosing and a 14-day gluten challenge included 17 receiving ANB033 and 15 placebo. At Week 6, the least-squares mean change in villus-height-to-crypt-depth ratio was -0.17 with ANB033 and -0.66 with placebo, a +0.49 placebo-adjusted delta (p=0.009). Intraepithelial lymphocyte density changed -1.20 versus +18.83, a -20.03 placebo-adjusted delta (p<0.001).

During the challenge, symptoms did not increase in the ANB033 group; total symptom score differences were statistically significant from days 1–7, and moderate-to-severe symptom differences through days 1–12. The safety analysis covered 34 participants: no serious adverse events occurred; any adverse event was reported in 16 of 18 ANB033 participants (88.9%) and 16 of 16 placebo participants (100%). First Tracks anticipates Cohort 2 celiac top-line data in Q1 2027 and ANB033 eosinophilic esophagitis data in Q3 2027; it anticipates starting a late-stage celiac trial in 2027.

How this balance works

Rhea-AI gives every point it takes from this document a weight. Minor counts 1, Moderate 3 and Major 9, so one Major point outweighs several Minor ones. The bar adds up the weights on each side, and when neither side holds more than 65% of the total the balance reads Mixed.

It reads the document as published, with the same rules for every company, and it does not look at what the market expected or at how the stock traded, so a point can be objectively good on a day the stock falls.

Rhea-AI Sentiment measures something else, the tone of the wording.

CD122 antagonist medical
"ANB033, an investigational, subcutaneously administered CD122 antagonist"
A CD122 antagonist is a drug that blocks the CD122 protein, a key part of the receptor that immune signals use to activate certain white blood cells. By preventing those signals, the drug can dial down immune activity—useful for treating autoimmune diseases, preventing transplant rejection, or altering anti‑cancer immune responses. Investors care because such a compound’s effectiveness and side‑effect profile drive clinical trial success and commercial potential, like adjusting a thermostat to change a room’s temperature.

"least squares (LS) mean change from baseline"
Least squares (LS) mean is an adjusted average calculated from a statistical model that removes the influence of other factors so groups can be compared on a level playing field. Think of it as the average outcome you would expect if everyone being compared had the same mix of background characteristics, like age or starting size. For investors, LS means matter because they give a fairer view of performance or effect sizes in studies and reports, helping avoid misleading conclusions driven by imbalances in the data.

SAN DIEGO, CA — Oct. 12, 2026 — First Tracks Biotherapeutics, Inc. (Nasdaq: TRAX), a clinical-stage biotechnology company advancing antibody therapeutics that modulate immune pathways implicated in autoimmune and inflammatory diseases, today announced positive top-line results from Cohort 1 (gluten challenge) of ANB033’s Phase 1b trial in celiac disease (CeD). ANB033, an investigational, subcutaneously administered CD122 antagonist, is designed to inhibit the signaling of both IL-15 and IL-2 on immune cells.

ANB033 demonstrated a statistically significant difference compared to placebo across key histologic endpoints, including least squares (LS) mean change in villus height-to-crypt depth (Vh:Cd) ratio, intraepithelial lymphocyte (IEL) density, and VCIEL composite scale, as well as symptom endpoints using the Celiac Disease Symptom Diary (CDSD).

In the context of an intense gluten challenge, treatment with ANB033 not only prevented an increase in IEL density, but also protected against gluten-induced intestinal damage with changes in Vh:Cd ratio remaining below the threshold considered clinically meaningful.

“For people living with celiac disease, trace exposure to gluten can cause intestinal damage and debilitating symptoms despite general adherence to a gluten-free diet. After treatment with ANB033, patients underwent a 14-day gluten challenge consuming the equivalent of two slices of bread a day. ANB033 achieved a statistically significant and clinically meaningful protective effect against gluten-induced intestinal damage and symptoms, while maintaining a favorable safety profile,” said Daniel Faga, president and chief executive officer. “We look forward to continuing to work with regulators, clinicians and the patient community as we advance ANB033 into late-stage development in 2027.”

The randomized, placebo-controlled Phase 1b trial in CeD is evaluating subcutaneous ANB033 administered across two cohorts: Cohort 1, a 6-week study assessing prevention of gluten-induced intestinal damage and symptoms after a 14-day gluten challenge, and Cohort 2, assessing mucosal healing in patients with a broad range of histologic and symptom severity through 12 weeks.

Cohort 1 evaluated 32 patients (17 on ANB033 and 15 on placebo) who completed dosing and the gluten challenge at Week 6. Patients received three subcutaneously administered loading doses of ANB033 or placebo at Week 0, Week 2, and Week 4, followed by a 14-day gluten challenge (6g daily, the equivalent of approximately two slices of bread,

administered by two gluten cookies). The trial assessed safety and tolerability as well as key histologic and symptom endpoints used to measure clinical benefit with a primary analysis at Week 6.

Key Efficacy Endpoints Were Achieved in Cohort 1

ANB033 achieved the key histologic efficacy endpoints of LS mean change from baseline in Vh:Cd ratio, IEL density, and VCIEL vs. placebo at Week 6.

Using the Celiac Disease Symptom Diary (CDSD), symptoms were assessed daily through the entire 14-day gluten challenge. The CDSD is a validated, patient-reported outcome measure that assesses the frequency and severity of five key celiac disease symptoms which are abdominal pain, bloating, nausea, diarrhea and tiredness.

Symptom findings during the gluten challenge were as follows –

•
Placebo patients experienced a marked increase in symptoms consistent with an intense gluten challenge
•
ANB033 prevented an increase in symptoms throughout the entire gluten challenge
•
47% of placebo patients and only 6% of ANB033 patients took permitted medications to mitigate their GI symptoms
•
Lower CDSD score on ANB033 vs. placebo, a statistically significant difference from the first day through the seventh day
•
Lower moderate-to-severe symptoms on ANB033 vs. placebo, a statistically significant difference from the first day through the twelfth day

“It is remarkable to see, for the first time, both statistically significant prevention of symptoms and Vh:Cd benefit in the same study. Equally impressive, ANB033 showed changes from baseline that remained below the threshold considered clinically meaningful in an intense gluten challenge,” said Benjamin Lebwohl, M.D., MS, professor of Medicine and Epidemiology at NY-Presbyterian/Columbia University Irving Medical Center and director of Clinical Research at the Celiac Disease Center at Columbia University. “ANB033 takes a novel approach to treating celiac disease by targeting CD122 and blocking both IL-15 and IL-2 signaling, two key drivers of inflammation that damage the intestine, with IL-2 closely linked to symptom development. For the largest autoimmune disease without an approved therapeutic option, these findings are an important advance for the celiac disease community.”

Safety analysis set includes two additional participants who were randomized in Cohort 1 but discontinued the study prior to initiating the gluten challenge, were therefore unevaluable for gluten challenge efficacy analysis and did not undergo the second biopsy at Week 6. The one participant on ANB033 discontinued due to a decision not to undergo the gluten challenge.

Next Steps for ANB033

First Tracks Biotherapeutics will continue to engage with regulatory agencies, including the U.S. FDA, which has already granted ANB033 Fast Track designation, to discuss and align on the next phase of development for ANB033 in CeD. We anticipate starting a late-stage clinical trial in CeD in 2027.

Cohort 2 of ANB033’s Phase 1b trial in CeD, which is assessing mucosal healing in patients with a broad range of histologic (Vh:Cd ratio ≤ 2.5) and symptom severity through 12 weeks, is on track to complete enrollment imminently, with top-line data anticipated in Q1 2027.

A Phase 1b cohort in eosinophilic esophagitis (EoE) is enrolling 50 patients with top-line data anticipated in Q3 2027. Additionally, we are also on track to initiate Phase 2 development in vitiligo and one additional indication in H1 2027.

“The consistency of the histologic and symptom findings strengthens our confidence in the underlying biology and supports continued development of ANB033 in celiac disease and other conditions driven by this pathway, including eosinophilic esophagitis and vitiligo,” added Paul Lizzul, M.D., Ph.D., chief medical officer. “We are deeply grateful to the patients, investigators and study teams who made this important milestone possible and look forward to presenting these results at ICDS in Melbourne in November.”

First Tracks Biotherapeutics Investor Call

Management will host an investor call and live webcast, with an accompanying slide presentation, to review the Cohort 1 results today, Monday, Oct. 12, 2026, at 8:30 a.m. ET / 5:30 a.m. PT. About ANB033

ANB033, a potentially best-in-class CD122 antagonist, targets CD122, the common beta subunit of the IL-15 and IL-2 receptors, to reduce subsets of CD8+ and CD4+ T cells, NK cells, as well as ILC2s. Antagonizing CD122 has the potential to achieve and maintain remission of inflammation through the reduction of disease-causing cytolytic CD8 T cell subsets, including IELs and NK cells and reducing inflammatory cytokine secretion by activated CD4+ Th1 and Th2 cells, as well as ILC2 cells.
