ADAPTIN BIO, INC. (APTN) announced that enrollment has opened for a Phase 1, open-label, dose-escalation clinical trial of its proprietary BRiTE (Brain Bispecific T cell Engager) therapeutic APTN-101 as a treatment for EGFRvIII-positive glioblastoma, in collaboration with Duke University.
The study will enroll up to 15 adult patients with World Health Organization Grade IV Malignant Glioma. The primary objective is to evaluate the safety profile and maximum tolerated dose, with the primary endpoint being the proportion of patients with a dose limiting toxicity at each dose level. Secondary endpoints include pharmacokinetics and objective response rate, and exploratory endpoints include cytokine changes, anti-BRiTE antibodies, overall survival and progression free survival.
In preclinical models, APTN-101 increased brain distribution of an EGFRvIII T-cell engager by more than 7-fold and completely eradicated EGFRvIII glioblastoma tumors in 70%–80% of mice, while showing an excellent safety profile. The company highlights the significant unmet need in glioblastoma, where median survival is 12–18 months and about 5% of patients survive beyond five years.
Adaptin Bio, Inc. reported that its common shares have been approved for quotation on the OTCQB Venture Market under the ticker symbol APTN. The company expects trading to begin in approximately 6–8 weeks and plans to share any material updates as they occur.
