Clinical and regulatory milestones have often driven strong upside for PRAX, though at least one major positive trial readout previously saw a negative price reaction, showing occasional sell-the-news behavior.

Recent Company History

Over the past two years, PRAX has repeatedly reported positive clinical milestones across its CNS pipeline. On Sep 3, 2024, positive EMBOLD data for relutrigine in SCN2A/SCN8A DEEs preceded further detailed topline results on Sep 3, 2024. The Phase 2 RADIANT data for vormatrigine on Aug 4, 2025 showed strong seizure reductions but saw a negative reaction. By Oct 16, 2025, best-in-disease Phase 3 Essential3 results for ulixacaltamide triggered a large rally, and the Mar 30, 2026 NDA acceptance for relutrigine also produced a solid gain. Today’s AAN 2026 Essential3 and pipeline showcase fits this pattern of meaningful clinical updates.

Key Terms

breakthrough therapy designation, phase 3, randomized withdrawal study, digital biomarker, +4 more
8 terms
breakthrough therapy designation regulatory
"ulixacaltamide has received FDA Breakthrough Therapy Designation, with data selected..."
A breakthrough therapy designation is a regulatory fast-track given to a drug or treatment that shows early signs of providing a major improvement over existing options for a serious condition. Think of it as a VIP lane that can speed up development and more intensive guidance from regulators, which matters to investors because it can shorten time to market, reduce development risk and potentially increase a company’s value — though it does not guarantee approval.
phase 3 clinical
"Essential3 is the first positive Phase 3 program in essential tremor; ulixacaltamide..."
Phase 3 is the late-stage clinical testing step for a new drug or medical treatment, where the product is given to large groups of patients to confirm effectiveness, monitor side effects, and compare it to standard care. Successful Phase 3 results are often the final scientific hurdle before regulators decide on approval and market launch—like passing a final exam before graduation—and can sharply change a company's valuation and future revenue prospects.
randomized withdrawal study clinical
"Results from Essential3 Study 2 (Randomized Withdrawal Study)"
A randomized withdrawal study is a clinical trial where all participants initially receive the experimental treatment, and those who improve are then randomly assigned either to continue the treatment or to switch to an inactive pill or stop treatment so researchers can see whether benefits persist. Investors care because this design shows durability of effect and how quickly symptoms return after stopping, which affects regulatory decisions, prescribing guidance, patient demand and long-term sales — like removing a coach to see if performance holds up on its own.
digital biomarker medical
"regulatory-grade Digital Biomarker Pipeline for the Essential3 Program"
A digital biomarker is measurable health information captured by sensors, apps, or wearable devices—such as heart rate patterns, walking speed, or sleep activity—that acts like a dashboard light indicating a person’s physical or mental condition. Investors care because these signals can speed drug development, enable new monitoring services, and create recurring data-driven revenue, while also carrying regulatory and validation risks that affect commercial and clinical value.
antisense oligonucleotide medical
"Complementary Antisense Oligonucleotide Treatment and Precision Sodium Channel Modulation..."
An antisense oligonucleotide is a small piece of synthetic genetic material designed to attach to specific molecules in the body’s cells, effectively blocking or modifying how genes are expressed. This technology is important because it can be used to develop targeted treatments for certain diseases, which may influence the value of biotech companies and the broader healthcare sector. Its development reflects advances in personalized medicine and gene-based therapies.
double-blind clinical
"A Phase 3, Randomized, Multi-center, Double-blind, Placebo-controlled Clinical Trial..."
A double-blind process means that neither the people conducting an activity nor the people involved know certain key details, such as who is receiving a treatment or a placebo. This approach helps prevent bias from influencing the results, making the outcome more trustworthy. For investors, it ensures that decisions or judgments are based on unbiased information rather than preconceived opinions or expectations.
placebo-controlled clinical
"Double-blind, Placebo-controlled Clinical Trial to Evaluate the Efficacy..."
"Placebo-controlled" describes a testing method where one group receives the actual treatment or intervention, while another group receives a harmless, inactive version called a placebo. This approach helps determine whether the real treatment has genuine effects beyond psychological expectations. For investors, understanding this ensures confidence that reported benefits are real and not influenced by bias or false perceptions.
developmental and epileptic encephalopathies medical
"Relutrigine in Participants with Developmental and Epileptic Encephalopathies..."
Developmental and epileptic encephalopathies are a group of severe brain disorders that begin early in life, causing significant challenges with learning, development, and seizures. These conditions can limit a child's ability to grow and function normally, often requiring ongoing medical care. For investors, understanding these disorders highlights areas of medical research and treatment development that could lead to new therapies and market opportunities.

Ulixacaltamide has received Breakthrough Therapy Designation from the FDA and is the most advanced program within Praxis’ Cerebrum™ small molecule platform.

About Vormatrigine
Vormatrigine is a next-generation, functionally selective small molecule targeting the hyperexcitable state of sodium-channels in the brain that is currently being developed as a once daily, oral treatment for adult focal onset seizures and generalized epilepsy. Preclinical data demonstrates vormatrigine is differentiated from standard of care, with the potential to be best-in-class for focal epilepsy. In vitro, vormatrigine has demonstrated superior selectivity for disease-state NaV channel hyperexcitability. In vivo studies of vormatrigine have demonstrated unprecedented potency in the maximal electroshock seizure (MES) model, a highly predictive translational model for efficacy in focal epilepsy. Data from patients in the RADIANT study demonstrated a robust seizure reduction and generally safe and well tolerated profile. To learn more about the POWER1 and POWER2 studies, please visit POWER studies.

About Relutrigine

Relutrigine is a first-in-class small molecule in development for the treatment of developmental and epileptic encephalopathies (DEEs) as a preferential inhibitor of persistent sodium current, shown to be a key driver of seizure symptoms in severe DEEs. Relutrigine’s mechanism of precision sodium channel (NaV) modulation is consistent with superior selectivity for disease-state NaV channel hyperexcitability. In vivo studies of relutrigine have demonstrated dose-dependent inhibition of seizures up to complete control of seizure activity in SCN2A, SCN8A and other DEE mouse models. Relutrigine has been generally well-tolerated in three Phase 1 studies and has demonstrated biomarker changes indicative of NaV channel modulation. Data from cohort 1 of the Phase 2 EMBOLD study demonstrated a well-tolerated, robust, short- and long-term improvement in motor seizures in a heavily pre-treated population, alongside maintained seizure freedom in some patients with SCN2A- and SCN8A-DEE. Relutrigine has received Orphan Drug Designation (ODD) and Rare Pediatric Disease Designation from the FDA for the treatment of SCN2A-DEE, SCN8A-DEE and Dravet syndrome; as well as Breakthrough Therapy Designation (BTD), and ODD from the European Medicines Agency for the treatment of SCN2A-DEE and SCN8A-DEE. To learn more about the EMERALD study, please visit Emerald | Resilience Studies.

About Elsunersen

Elsunersen is an antisense oligonucleotide (ASO) designed to selectively decrease SCN2A gene expression, directly targeting the underlying cause of early-seizure-onset SCN2A-DEE to treat seizures and other symptoms in patients with gain-of-function SCN2A mutations. In vitro studies of elsunersen have demonstrated reduction in both SCN2A gene expression and protein levels. In vivo, elsunersen has demonstrated significant, dose-dependent reduction in seizures, improvement in behavioral and locomotor activity and increased survival in SCN2A mouse models, with potential to be the first disease-modifying treatment for SCN2A-DEE. Elsunersen has received ODD and RPDD from the FDA, and ODD and PRIME designations from the European Medicines Agency for the treatment of SCN2A-DEE. The elsunersen program is ongoing under a collaboration with Ionis Pharmaceuticals, Inc., and RogCon, Inc. To learn more about the EMBRAVE3 study, please visit Embrave | Resilience Studies.
